GLP-1, an incretin hormone of the enteroinsular axis with insulinotropic and glucagonostatic activity, is secreted after nutrient ingestion. GLP-1 is mainly produced by intestinal L-cells in the lower gastrointestinal tract (GIT); simple carbohydrates are absorbed in the upper GIT and alpha-glucosidase inhibition leads to augmented and prolonged GLP-1 release in normal subjects. In a cross-over study, 100 mg acarbose or placebo was administered simultaneously with 100 g sucrose to 11 hyperglycaemic Type 2 diabetic patients poorly controlled with diet and sulphonylureas. Plasma levels of GLP-1, insulin, C-peptide, glugacon, GIP, glucose and H2-exhalation were measured over 6 h. Differences in the integrated responses over the observation period were evaluated by repeated measurement analysis of variance with fasting values used as covariates. With acarbose, sucrose reached the colon 60-90 min after ingestion as indicated by a significant increment in breath hydrogen exhalation (p = 0.005). After an early GLP-1 increment 15 min after sucrose under both conditions, GLP-1 release was prolonged in the acarbose group (p = 0.001; significant from 210 to 360 min.). Initially (0-150 min), glucose (p = 0.001), insulin (p = 0.001), and GIP (p < 0.001) were suppressed by acarbose, whereas later there were no significant differences. Glucagon levels were higher with acarbose in the last 3 h of the 6 h observation period (p = 0.02). We conclude that in hyperglycaemic Type 2 diabetic patients, ingestion of acarbose with a sucrose load leads to elevated and prolonged GLP-1 release.  相似文献

Presents a novel approach for automating the timing design of interfaces between VLSI chips in microcomputer systems. The Prolog-based expert system, called TDS (for timing design system), incorporates the heuristic knowledge of the hardware designer. TDS is a rule-based system that interprets the specification sheets of VLSI chips and can synthesize, diagnose, and verify timing charts at the expert's level. The system uses a functional model based on timing specifications, not the structural information. TDS can model other interfaces that are based on timing specifications, such as standard bus interfaces  相似文献

The aim of this study was to prepare magnetic beads that could be used for the removal of heavy‐metal ions from synthetic solutions. Magnetic poly(ethylene glycol dimethacrylate–1‐vinyl‐1,2,4‐triazole) [m‐poly(EGDMA–VTAZ)] beads were produced by suspension polymerization in the presence of a magnetite Fe3O4 nanopowder. The specific surface area of the m‐poly(EGDMA–VTAZ) beads was 74.8 m2/g with a diameter range of 150–200 μm, and the swelling ratio was 84%. The average Fe3O4 content of the resulting m‐poly(EGDMA–VTAZ) beads was 14.8%. The maximum binding capacities of the m‐poly(EGDMA–VTAZ) beads from aquous solution were 284.3 mg/g for Hg2+, 193.8 mg/g for Pb2+, 151.5 mg/g for Cu2+, 128.1 mg/g for Cd2+, and 99.4 mg/g for Zn2+. The affinity order on a mass basis was Hg2+ > Pb2+ > Cu2+ > Cd2+> Zn2+. The binding capacities from synthetic waste water were 178.1 mg/g for Hg2+, 132.4 mg/g for Pb2+, 83.5 mg/g for Cu2+, 54.1 mg/g for Cd2+, and 32.4 mg/g for Zn2+. The magnetic beads could be regenerated (up to ca. 97%) by a treatment with 0.1M HNO3. BACKGROUND: Medicinal plants commonly used and traded in Turkey were monitored for their cadmium and selected micronutrient (copper, iron, manganese and zinc) content. RESULTS: Cadmium was not detected in any sample of licorice extract, linden flowers or nettle leaves. However, in the rest of the samples, cadmium was found in the range of 7–126 µg kg?1, with the highest concentration in chamomile leaf. Of all the monitored trace elements, the highest concentration found was 520 mg kg?1 iron in zahter. The highest copper, manganese and zinc concentrations were found in rose hips (24 mg kg?1), Turkish oregano (58 mg kg?1) and zahter (50 mg kg?1), respectively. CONCLUSION: The results of the present study reveal that cadmium and trace element concentrations in medicinal plants commonly used as traditional medicines in Turkey are well below the critical levels. Scope: Cancer cachexia is characterized by muscle and adipose tissue wasting caused partly by chronic, systemic inflammation. Conjugated linoleic acids (CLAs) are a group of fatty acids with various properties including anti‐inflammatory cis9, trans11 (c9t11)‐CLA and lipid‐mobilizing trans10, cis12 (t10c12)‐CLA. The purpose of this study was to test whether dietary supplementation of a c9t11‐CLA‐rich oil (6:1 c9t11:t10c12) could attenuate wasting of muscle and adipose tissue in colon‐26 adenocarcinoma‐induced cachexia in mice. Methods and results: Loss of body weight, muscle and adipose tissue mass caused by tumors were not rescued by supplementation with the c9t11‐CLA‐rich oil. In quadriceps muscle, c9t11‐CLA‐rich oil exacerbated tumor‐induced gene expression of inflammatory markers tumor necrosis factor‐α, IL‐6 receptor and the E3 ligase MuRF‐1 involved in muscle proteolysis. In epididymal adipose tissue, tumor‐driven delipidation and atrophy was aggravated by the c9,t11‐CLA‐rich oil, demonstrated by further reduced adipocyte size and lower adiponectin expression. However, expression of inflammatory cytokines and macrophage markers were not altered by tumors, or CLA supplementation. Conclusion: These data suggest that addition of c9t11‐CLA‐rich oil (0.6% c9t11, 0.1% t10c12) in diet did not ameliorate wasting in mice with cancer cachexia. Instead, it increased expression of inflammatory markers in the muscle and increased adipose delipidation.  相似文献

The use of oxidoreductases (EC1) in non-conventional reaction media has been increasingly explored. In particular, deep eutectic solvents (DESs) have emerged as a novel class of solvents. Herein, an in-depth study of bioreduction with an alcohol dehydrogenase (ADH) in the DES glyceline is presented. The activity and stability of ADH in mixtures of glyceline/water with varying water contents were measured. Furthermore, the thermodynamic water activity and viscosity of mixtures of glyceline/water have been determined. For a better understanding of the observations, molecular dynamics simulations were performed to quantify the molecular flexibility, hydration layer, and intraprotein hydrogen bonds of ADH. The behavior of the enzyme in DESs follows the classic dependence of water activity (aW) in non-conventional media. At low aW values (<0.2), ADH does not show any activity; at higher aW values, the activity was still lower than that in pure water due to the high viscosities of the DES. These findings could be further explained by increased enzyme flexibility with increasing water content.  相似文献

In the present study, 17 wt % TiN reinforced α-β SiAlON composites were sintered at low temperature by susceptor-assisted microwave heating. The effect of TiN addition on dielectrical properties of starting powders, as well as the influence of sintering temperature on phase evolution, microstructure development and mechanical properties of α/β-SiAlON-TiN composites were investigated. The obtained results showed that TiN addition increased the microwave absorbing properties which is reflected in the peak sintering temperature. Thus, the α:β ratio decreased and mechanical properties were improved, especially the fracture toughness of the composites. Furthermore, an estimate of energy consumption during microwave assisted sintering at the laboratory scale is presented. As a result, the highest values for relative density (97.1%), Vickers hardness (13.35 ± 0.47 GPa), and fracture toughness (7.52 ± 0.54 MPa m1/2) were obtained by microwave sintering for 30 min at 1300 °C.  相似文献

Chemical transmission lines, petroleum and natural gas lines, pressure vessels, and pipes used in thermal facilities are expected to maintain their mechanical properties for many years without being damaged and not to be corroded in working conditions. The composite materials are the right candidate for these harsh conditions due to their superior properties. Reinforcement of nanoadditives to composite materials improves both the mechanical properties and the resistance to environmental conditions, thereby increasing the lifetime. In this study, multi-walled carbon nanotube (MWCNT) reinforced [±?55°] carbon fiber/epoxy composite pipes produced with filament wound method were used. It was hydrothermally aged in 80 °C distilled water for 1, 2, 3 weeks in order to examine the effect of environmental conditions. In order to investigate its resistance against loads that may occur in working conditions, ring tensile tests (ASTM D 2290–16 procedure A), and low-velocity impact tests at 5, 10, 15 J, energy levels were carried out. The effect of hydrothermal aging on neat and MWCNT added epoxy composite had been examined by considering the aging period. Consequently, the impact resistance of neat and MWCNT added samples decreased with the aging process. Besides, tangential tensile strength loss was 17% in MWCNT reinforced sample and 13% in the neat sample.
